چکیده

During determination of the oxygen capacity of blood by the Van Slyke method (1), the following adaptations were made in order to reduce errors: (1) Blood was equilibrated with water-saturated air in a siliconized tonometer. (2) The determination of the oxygen content of the sample was carried out immediately after equilibration. (3) Triton was used instead of saponin in the ferricyanide reagent. (4) Special care was taken in the preparation and storage of the dithionite reagent, which should be prepared fresh daily and stored in the dark at 4#{176}. The hemoglobin content and oxygen capacity, measured by the modified technic, were compared in a series of 36 samples of human blood.

Manometric Determination of Hemoglobin by the Oxygen Capacity Method

Since Van Slyke (1) originally described a gasometric method for the determination of hemogIobin by analysis of the oxygen-combining power of blood, it has been the practice to saturate the blood with air at atmospheric pressure in a separate vessel and to transfer a sample of the aerated blood to the gas analyzing apparatus. A method is described here which requires less time, material, and ap...

Gasometric Determination of Hemoglobin by the Carbon Monoxide Capacity Method

The method here described is based on the capacity of hemoglobin to combine with the same maximum volume of CO as of 02. The blood sample, which may vary from 0.1 to 2.0 cc., is saturated with CO in the 50 cc. chamber of the manometric blood gas apparatus of Van Slyke and Neil1 (1, 2). Approximately 2 cc. of carbon monoxide are introduced with the blood, and the chamber is evacuated and shaken....

The Iron Content of the Whole Blood of Normal Individuals

During t.he course of an investigation involving the determination of blood iron values in normal subjects, it became evident that t.he results were consistently higher than the figures recently reported for human blood by Murphy, Lynch, and Howard (1) and Reich and Tiedemann (2). Furt, hermore, hemoglobin values calculat,ed on the basis of their iron determinations differed considerably from va...

Spectrophotometric determination of blood oxygen.

Gasometric methods for estimating blood oxygen are time-consuming. This consideration often limits the number of observations which can be made in the course of experiments requiring oxygen determinations. In many cases it is an arteriovenous difference which is particularly desired, as in the measurement of cardiac output by the Fick principle, or the estimation of oxygen consumption by human ...

The Refractometric Determination of Hemoglobin

The standard method of determining hemoglobin concentration in blood is based on a measurement of oxygen capacity and Htifner’s factor, 1.34 cc. of oxygen per gm. of hemoglobin. Unfortunately, the Van Slyke and the Haldane machines do not give the same results for the oxygen capacity. The difference is over 10 per cent of the total, and there is no agreement as to which method is correct. The c...
